You can get the virus, adware, trojan etc. all together called malwares for malicious softwares from web sites, downloads, or files you might share.
If you share files between the two machines, it is possible to infect the desktop PC.
Most malware nowadays are distributed through script and launched using social engineering - talking the recipient of the attack to open an e-mail or click a link, approve an install or download a package.
As far as malware is concerned your router, is just like any other router on the Internet - to most malware - transparent. Malware does not care in general about routers.

The Desk top should be fine. My dad has the same type of router and the only way to get a virus from the router is if you get one on the computer that is giving off the router signal.
Not ''automatically''. I suppose there's a way someone could arrange to pass an infection, but one of you would have to do so intentionally--which obviously is not what you want.
